pLenti CMVtight Series

These vectors are compatible with Tet-On Advanced cell lines (Clontech). In the absence of the reverse tetracycline transactivator 3 (rtTA3), they should not show any expression, although low expression has been detected (result not shown). Cell lines can be made Tet-On Advanced using the pLenti CMV rtTA3 Blast (w756-1) or pLenti CMV rtTA3 Hygro (w785-1) plasmid.

NOTE: If the vector pLenti CMV rtTA3 Blast (w756-1) was used to make the Tet-On cell line, this vector can not be selected for because the cell line will be already resistant to blasticidin. The rtTA3 element should be expressed under a different drug selection such as hygromycin using the pLenti CMV rtTA3 Hygro (w785-1) vector.
